Abstract: At higher temperatures, lettuce seeds may not germinate, resulting in problems for crop establishment in the field and production. This study aimed to evaluate the maternal effect for thermotolerance on lettuce seeds and find enzymatic markers. We used the thermoinhibition tolerant cultivar ‘Everglades’, the sensitive ‘Verônica’, their reciprocal hybrids, F1 and F2 generation. Seeds were submitted to germination and enzymatic tests (catalase, superoxide dismutase, ascorbate peroxidase, and endo- β-mannanase). Germination (first and final counting) and speed index were compared through Tukey’s test. ‘Everglades’ and its reciprocal hybrids, have not been influenced by temperature regarding germination compared to the other treatments. Also, the maternal effect was observed for the F1 generation. Higher temperatures interfere with the activity of all enzymes evaluated, consequently in the physiological parameters. However, superoxide dismutase and endo- β-mannanase have shown direct and more expressive correlation with changes on the physiological parameters compared to the other enzymes. Thus, it is possible to conclude that those enzymes can be used as markers for thermotolerant genotype selection.

Resumo: Sob altas temperaturas, sementes de alface não germinam, resultando em problemas para o estabelecimento da cultura em condição de campo. Esse trabalho analisou a possibilidade de presença de efeito maternal na termotolerância em sementes de alface e buscou marcadores enzimáticos para tal. Usaram-se as cultivares Everglades (tolerante) e ‘Verônica’ (sensível), bem como seus híbridos recíprocos e a geração F2. As sementes foram analisadas quanto à germinação, bem como atividade enzimática (catalase, superóxido dismutase, ascorbato peroxidase e endo-β-mananase). O percentual de germinação; (primeira contagem e final), bem como a velocidade (IVG) foram analisados por meio de ANOVA e teste de Tukey. A cultivar Everglades e seus híbridos recíprocos não foram influenciados pela germinação em alta temperatura quando comparados com os outros tratamentos. Além disso, o efeito maternal foi observado na geração F1. Altas temperaturas interferem na atividade de todas as enzimas analisadas nesta pesquisa, consequentemente também nos parâmetros fisiológicos. Contudo, a superóxido dismutase e a endo-β-mananase tiveram suas variações altamente correlacionadas com as mudanças nos parâmetros fisiológicos. O presente trabalho evidenciou o efeito materno para a termotolerância em sementes de alface, bem como o uso da superóxido dismutase e endo-β-mananase como marcadores enzimáticos para tal característica.

Abstract The objective of this work was to evaluate the occurrence of the maternal effect on thermotolerance in lettuce (Lactuca sativa) seeds, as well as to find an enzymatic marker for this trait in lettuce seedlings. Seeds from the Everglades thermotolerant cultivar, the Verônica susceptible cultivar, and their reciprocal hybrids were used. For each cultivar and reciprocal hybrid, germination percentage and vigor (germination speed index) tests were carried out at 20 and 32°C. Thermotolerance was defined as the capacity to germinate at 32°C. To obtain markers that could be associated with thermotolerance, enzymatic tests were performed. The bands of the electrophoretic enzymatic extracts were quantified using the ImageJ software. The hybrids behaved similarly to their female genitors. When cultivar Verônica was the female genitor, hybrid seeds did not germinate at high temperatures and showed a lower expression of the esterase enzyme. However, when the female genitor was the Everglades cultivar, hybrids showed a high germination at high temperatures and a greater esterase expression. Therefore, there is a maternal effect on tolerance to thermoinhibition in lettuce seeds, and the esterase enzyme can be used as a selective marker to identify homozygous progenies for this trait.

Resumo O objetivo deste trabalho foi avaliar a ocorrência do efeito materno na tolerância à termoinibição em sementes de alface (Lactuca sativa), bem como encontrar um marcador enzimático para esta característica em mudas de alface. Foram utilizadas sementes da cultivar termotolerante Everglades, da cultivar suscetível Verônica e de seus híbridos recíprocos. Para cada cultivar e híbrido recíproco, foram conduzidos testes de germinação e vigor (índice de velocidade de germinação) a 20 e 32°C. A termotolerância foi definida pela capacidade de germinação a 32°C. Para encontrar marcadores que pudessem ser associados com a termotolerância, foram realizados testes enzimáticos. As bandas eletroforéticas dos extratos enzimáticos foram quantificadas por meio do programa ImageJ. Os híbridos apresentaram comportamento semelhante ao dos seus genitores femininos. Quando a cultivar Verônica foi o genitor feminino, as sementes dos híbridos não germinaram sob alta temperatura e apresentaram menor expressão da enzima esterase. Porém, quando a cultivar Everglades foi o genitor feminino, os híbridos apresentaram alta germinação sob temperatura elevada e maior expressão da esterase. Portanto, há efeito materno na tolerância à termoinibição em sementes de alface, e a enzima esterase apresenta potencial como marcador para identificar progênies homozigotas para essa característica.

Resumo: Vários estudos têm utilizado com sucesso o polietilenoglicol (PEG) e/ou ácido abscísico (ABA) para induzir a tolerância à dessecação em sementes germinadas de espécies ortodoxas, melhorando nosso entendimento dos mecanismos que fundamentam essa característica. Nesse contexto, tratamentos com PEG e ABA foram testados para avaliar sua eficiência em reduzir a sensibilidade à dessecação e aumentar o período de armazenamento das sementes de Magnolia ovata. A secagem das sementes a 10% de teor de água não afetou significativamente a germinação, independentemente de terem sido previamente tratadas ou não com ABA. No entanto, sementes tratadas com PEG apresentaram menor viabilidade. Após o armazenamento, as sementes tratadas com ABA e dessecadas a 5% de teor de água, apresentaram maiores taxas de germinação, sendo que o tratamento com ABA 100 µM proporcionou a maior capacidade de armazenamento. Não foram observadas alterações na atividade da catalase, peroxidase ou superóxido dismutase ou na abundância de proteínas totais ou resistentes ao calor.

Abstract: Several studies have successfully used polyethylene glycol (PEG) and/or abscisic acid (ABA) to reinduce desiccation tolerance in germinated seeds of orthodox species, improving our understanding of the mechanisms that underlie this trait. In this context, treatments with PEG and ABA were tested to assess their efficiency in reducing the sensitivity to desiccation and increasing the storage period of Magnolia ovata seeds. The drying of seeds to 10% water did not significantly affect germination, nor was germination impaired in seeds that were dried and treated with ABA. However, seeds treated with PEG had lower viability. After storage, ABA-treated seeds with as low as 5% water content (WC) had higher germination rates, the treatment with 100 µM ABA providing the greatest storage capacity. No changes were observed in the activity of catalase, peroxidase, or superoxide dismutase or in the abundance of total or heat-resistance proteins.

Resumo: Com o crescimento da produção de forrageiras, a demanda por sementes de alta qualidade aumenta. O condicionamento fisiológico vem como técnica para produzir sementes de alta qualidade e tolerância à estresses. O objetivo deste trabalho foi avaliar o efeito de agentes condicionantes sobre a germinação de sementes e crescimento de plântulas de Urochloa ruziziensis em condições de estresse hídrico e salino. Foi estabelecido um experimento em delineamento inteiramente casualizado testando o efeito de seisagentes condicionantes e três condições de germinação. Foram avaliados conteúdo de água, porcentagem e velocidade de germinação, viabilidade das sementes remanescentes do teste de germinação, comprimento da plântula, atividade do sistema antioxidante e a quantidade do malonaldeído. Melhor performance foi observada em sementes condicionadas submetidas ao déficit hídrico e salino do que aquelas não condicionadas. Um aumento significante foi observado na percentagem e velocidade de germinação de sementes, bem como no tamanho de raízes e caule. O nitroprussiato de sódio tem maior potencial para uso no condicionamento fisiológico de sementes de U. ruziziensis.

Abstract: As forage production increase, high-quality seeds demand follows. Priming have been reported as a technique for improve seed quality and stress tolerance. The aim of this study was to evaluate the priming agents effect on Urochloa ruziziensis seed germination and seedling establishment under water deficit and salt stress. A completely randomized experiment assessing six priming agents and three germination conditions were established. We evaluated: seed water content; germination percentage and speed index; viability of seeds remaining from the germination test; seedling length; antioxidant enzymatic activity; and malondialdehyde quantity. Seeds primed under water deficit and salt stress showed better physiological performance than dry seeds. There was a significant increase in seed germination percentage and speed and in seedling roots and shoots length. Sodium Nitroprusside shows significant potential for use in the physiological priming of U. ruziziensis seeds.

Abstract Some species of the Lauraceae family produce seeds that are generally sensitive to desiccation, which makes them difficult to store. The objective of this study was to characterize changes in seed quality of C. aschersoniana from two lots, as well in the physiological and cellular aspects during 12 months of storage. Seeds were stored with their original moisture content (MC) or after pre-drying to 35% MC in a cold chamber (5 °C) at a relative humidity of 40%. Seeds were sampled and tested at time 0, 3, 6 and 12 months of storage regarding to moisture content, germination and ultrastructural features. The seeds were dispersed with dormancy that was overcame by the cold storage condition and the reserves in undried seeds were partly consumed during storage. Both undried and pre-dried seeds remained viable for at least 12 months.

ABSTRACT The maturation process of Sesbania virgata seeds was investigated through fruits and seeds at different maturation stages, where they were described morphologically. For the seeds, fresh and dry mass and moisture were determined and structural analysis of the tegument under a light microscope was performed. Germination tests in the presence and absence of drying and the electrophoretic profile of heat resistant proteins were also performed. The results showed that the seeds reached physiological maturity at 44 days after anthesis (DAA), evidencing a secondary cell wall. The acquisition of desiccation tolerance was observed at 44 DAA, with the seeds responding to the drying treatment. Protein bands were observed from 32 DAA, which remained constant until the end of maturation. These results allowed the characterization of the development and acquisition of desiccation tolerance in S. virgata seeds.

RESUMO A compreensão da tolerância à dessecação é de grande interesse para a tecnologia de sementes, visto que tem aplicação prática no estabelecimento de estratégias para a conservação de sementes e produção de mudas. O presente trabalho objetivou estudar a perda da tolerância à dessecação durante a germinação de sementes das espécies arbóreas sucupira (Bowdichia virgilioides), pau-ferro (Libidibia ferrea), cedro (Cedrela fissilis), tamboril (Enterolobium contortisiliquun), ipê-roxo (Handroanthus impetiginosus) e pau-jacaré (Piptadenia gonoacantha). Sementes coletadas na região de Lavras - MG foram usadas para os experimentos, sendo estes realizados isoladamente para cada espécie. Primeiramente foi estabelecida a curva de embebição por meio da pesagem das sementes nas condições de germinação a intervalos regulares. Baseado nessa informação, as sementes foram postas para embeber por tempos determinados pela curva de embebição e submetidas à secagem até atingir o mesmo percentual de umidade encontrado antes da semeadura. Em seguida as sementes foram reidratadas e aquelas que continuaram a germinação e originaram plântulas normais foram consideradas tolerantes à dessecação. Cada espécie apresentou um padrão próprio quanto à perda da tolerância à dessecação, sendo que em todos os casos, não houve tolerância à dessecação após a protrusão da raiz primária. Contudo, foi observada em sementes de pau-jacaré uma redução mais drástica logo no início do processo germinativo. Nas condições do presente trabalho, observou-se perda da tolerância à dessecação nas primeiras etapas do processo germinativo em todas as espécies, o que pode ter implicações em trabalhos que envolvam o condicionamento fisiológico das sementes.

ABSTRACT Understanding the loss of desiccation tolerance is of great importance in seed technology for its implications in the development of strategies for seed conservation and seedling production. In the present work, the loss of desiccation tolerance was studied in seeds of tree species Bowdichia virgilioides, Libidibia ferrea, Cedrela fissilis, Enterolobium contortisiliquum, Handroanthus impetiginosus, and Piptadenia gonoacantha. The seeds were collected in the region of Lavras, MG, and subjected to desiccation experiments individually for each species. Imbibition curve was created for each species by measuring seed weight at regular intervals under germination conditions. Based on this information, the seeds were allowed to imbibe for a time inferred from the imbibition curve, and then allowed to dry until reaching the initial water content measured before the imbibition begun. Then, the seeds were rehydrated, and those that developed into normal seedlings were considered desiccation-tolerant. The loss of seed desiccation tolerance followed a specific pattern in each studied species, but none of the species was tolerant to desiccation after radicle protrusion. A drastic decrease in desiccation tolerance was observed in P. gonoacantha seeds at the beginning of the germination process. Under the conditions implemented in this study, the loss of desiccation tolerance in all species started at the beginning of the germination process. These results have implications in both theoretical studies (seed priming) and the practice (in seedling production).

Resumo: O conhecimento sobre estratégias de regeneração natural é importante devido a sua influência no ecossistema local. Objetivou-se estudar o comportamento do banco de sementes aéreo de P. dubium. Os frutos foram colhidos mensalmente antes e após a maturação das sementes em matrizes localizadas no município de Lavras - MG. Mensalmente, foram determinados o conteúdo de água, aquisição da tolerância a dessecação, viabilidade das sementes, manutenção da dormência e tempo médio de germinação. Verificou-se que a maturidade fisiológica da espécie ocorreu no mês de junho, quando foi observado baixo conteúdo de água, máxima matéria seca, e alta porcentagem de germinação. Verificou-se que não houve alteração na dormência, porcentagem de germinação e no perfil proteico de sementes durante o período em que permanecem no banco de sementes aéreo. A espécie P. dubium forma banco de sementes aéreo de curta serotinosidade, com frutos permanecendo presos à planta por 9 meses.

Abstract: Knowledge concerning natural regeneration strategies is important due to the effect of natural regeneration on the local ecosystem. The aim of this study was to examine the ecophysiological aspects of the Peltophorum dubium aerial seed bank. Fruits were harvested monthly, before and after seed maturation, from selected trees in the municipality of Lavras, MG, Brazil, for determination of moisture content, acquisition of tolerance to desiccation, seed viability, maintenance of dormancy, and mean germination time. Physiological maturity of the species was attained in June, when low moisture content, maximum dry matter, and high germination percentage were observed. We found that there was no change in dormancy, in germination percentage, or in the protein profile of seeds during the period they remained in the aerial seed bank. The P. dubium species creates an aerial seed bank of short serotiny, with fruits remaining attached to the tree for 9 months.

Resumo: Este trabalho teve-se como objetivo avaliar o efeito do condicionamento osmótico na germinação de sementes de Eucalyptus urophylla e do híbrido de E. grandis x E. urophylla em condições de estresse salino. Foram testados dois potenciais de condicionamento induzidos por PEG 6000 (-1,0 e -1,5 MPa) em dois períodos (1 e 3 dias). Após o condicionamento osmótico, as sementes foram colocadas para germinar sob estresse salino em solução de NaCl nos potenciais 0,0 (controle) -0,5, -0,75 e -1,0 MPa a 25 °C. Os parâmetros avaliados foram a germinação e o índice de velocidade de germinação (IVG). Houve diminuição da germinação e do IVG à medida que o potencial hídrico do meio germinativo tornou-se mais negativo, tanto para E. urophylla quanto para o híbrido, entretanto, o E. urophylla foi mais tolerante ao estresse salino, em comparação ao híbrido, apresentando maior germinação em todos os potenciais hídricos testados. O osmocondicionamento no potencial de -1,0 MPa por três dias foi o mais indicado tanto para E. urophylla quanto para E. urophylla x E. grandis, quando submetidos à germinação em solução salina a -1,0 MPa, demonstrando que esse tratamento foi eficiente em favorecer a tolerância ao estresse salino.

Abstract: The objective of this experiment was to evaluate the effect of priming on the germination of Eucalyptus urophylla and of hybrid E. urophylla × E. grandis seeds under salt stress. Two osmotic potentials (-1.0 and -1.5 MPa) were tested, using PEG 6000 for 1 and 3 days. After priming, seeds were germinated under salt stress in a NaCl solution at 0.0 (control), -0.5, -0.75 and -1.0 MPa potentials, at 25 °C. Seed germination and germination speed index decreased as the water potential of the germination medium decreased. However, E. urophylla was more tolerant to salt stress; it showed a higher germination percentage under all tested potentials, when compared to the hybrid. The osmotic conditioning at -1.0 MPa for three days was more effective when E. urophylla x E. grandis was germinated in a salt solution at -1.0 MPa, indicating that this treatment was more effective in inducing tolerance to salt stress.

O objetivo deste trabalho foi avaliar a sensibilidade à dessecação de sementes de Tapirira obtusa (Benth.) J. D. Mitchell provenientes de três ambientes e submetidas a duas velocidades de secagem. As sementes foram coletadas em área de campo rupestre, Cerrado e em mata ciliar, da região de Lavras, MG. As sementes foram submetidas à secagem com cloreto de magnésio (secagem lenta) ou sílica gel (secagem rápida), em ambiente fechado, até os graus de umidade de 40%, 30%, 20% e 10%, considerando como controle o percentual de germinação na umidade inicial em cada ambiente, a qual variou de 47% a 50%. Foram avaliados os percentuais de germinação e de plântulas normais e também o índice de velocidade de germinação. Nos três ambientes analisados, não houve efeito da secagem lenta sobre a germinação das sementes. Sementes oriundas do Cerrado apresentaram pequena redução da germinação, quando submetidas à secagem rápida. Sementes oriundas de áreas rupestres apresentaram aumento na germinação quando submetidas à secagem rápida. Sementes oriundas de mata ciliar não apresentaram redução na germinação, independente da velocidade de secagem. Os resultados sugerem que sementes de T. obtusa não apresentam sensibilidade à dessecação.

This study was aimed at evaluating the desiccation sensitivity in seeds of the tree Tapirira obtusa (Benth.) J. D. Mitchell collected from three different environments and subjected to two distinct drying speeds. Seeds were collected from a rocky area, in the "Cerrado", and in a riparian forest area, in the region of municipality of Lavras, State of Minas Gerais. The seeds were subjected to drying with magnesium chloride (slow drying) or silica gel (fast drying), into closed environment, until moisture contents of 40%, 30%, 20% and 10%, considering as control, the percentage of germination at the initial moisture content in each environment, which varied from 47% to 50%. Percentages of germination and normal seedlings as well as germination speed index were assessed. For the three environments studied, there was no effect of slow drying on seed germination. Seeds from area of Cerrado, however, have shown a slight reduction on germination when subjected to fast drying. Oppositely, seeds from rocky area had germination increased when subjected to fast drying. Seeds from riparian forest area had no reduction on germination percentage, independent of drying speed. Results suggest that seeds of T. obtusa are not sensitive to desiccation.

Passiflora alata, P. cincinnata e P. setacea são frutíferas de grande importância, no Brasil, representando grande potencial agronômico. Entretanto, apresentam dificuldades de propagação, relacionadas à dormência e armazenamento das sementes. Desta forma, o presente estudo objetivou avaliar os efeitos do período e métodos de armazenamento sobre a emergência de plântulas de Passiflora alata Curtis, P. cincinnata Mast e P. setacea D.C. Para cada espécie, as sementes foram armazenadas à sombra, em refrigerador (10°C) e sob condições de laboratório e estratificação a 10°C. Aos 0, 7, 60 e 120 dias após o armazenamento, para P. alata e P. cincinnata, e 0, 7, 90 e 120 dias, para P. setacea, 50 sementes, para cada tratamento, foram semeadas em substrato comercial, com 4 repetições. Os experimentos foram estabelecidos em D.I.C., em esquema fatorial 4x3 (período x método). Avaliou-se a emergência de plântulas, a cada três dias, até 90 dias após a semeadura. Para P. alata, a estratificação proporcionou aumento nos percentuais de emergência (11%), após 60 dias de armazenamento das sementes, e redução aos 120 dias, não havendo influência positiva dos demais tratamentos. Não houve influência do método de armazenamento em P. cincinnata, observando-se aumento na emergência de plântulas até 60 dias de armazenamento (18,5%) e redução após este período. Em P. setacea, a estratificação resultou em aumento linear na emergência das plântulas, em função do tempo de armazenamento. A conservação de sementes de P. alata e P. setacea tem maior eficiência sob estratificação a frio. Sementes de P. cincinnata, independentemente do método utilizado, não devem ser armazenadas por mais de 60 dias.

Passiflora alata, P. cincinnata, and P. setacea are very important species, in Brazil, with high agronomic potential. However, they present propagation difficulties, related to seed dormancy and storage. Thus, this study aimed to evaluate the effects of storage period and methods on the Passiflora alata Curtis, P. cincinnata Mast, and P. setacea D.C. seedlings emergence. For each species, seeds were stored at shadow, in a refrigerator (10ºC) and under laboratory conditions and stratification at 10ºC. At 0, 7, 60, and 120 days after storage, for P. alata and P. cincinnata, and 0, 7, 90, and 120 days, for P. setacea, 50 seeds were sown in a commercial substrate, with 4 replications. The experiments were randomly designed in a 4x3 factorial scheme (period x method). The seedlings emergence was evaluated every three days, up to 90 days after sowing. For P. alata, the stratification provided an increase on emergence percentages (11%), 60 days after seeds storage, and decrease at 120 days, with no positive influence of the other treatments. There was no influence of the storage method for P. cincinnata, but it was noticed an increase in seedlings emergence up to 60 days after storage (18,5%) and a decrease from that period on. For P. setacea, the stratification resulted in a linear increase in seedlings emergence, according to the storage period. The conservation of P. alata and P. setacea seed is more efficient under cold stratification. P. cincinnata seeds, regardless of method, should not be stored for more than 60 days.

Este trabalho teve como objetivo avaliar o efeito da espécie de Passiflora usada como porta-enxerto no pegamento e desenvolvimento da microenxertia interespecífica ex vitro e caracterizar anatomicamente a região em que ela é realizada. Ápices caulinares de Passiflora edulis f. flavicarpa foram enxertados em hipocótilo de porta-enxertos de P. alata, P. cincinnata, P. edulis e P. setacea. Foram avaliados o desenvolvimento da microenxertia, a ocorrência de brotações adventícias do porta-enxerto e o percentual de microenxertos com folhas expandidas. Os porta-enxertos foram submetidos à avaliação anatômica e histoquímica. O desenvolvimento dos microenxertos e o número de brotações adventícias foram maiores quando se utilizou porta-enxertos de P. edulis e menores com porta-enxertos de P. setacea e P. alata. Porta-enxertos de P. edulis, P. cincinnata, P. alata e P. setacea proporcionaram, respectivamente, 30,3, 10, 1,6 e 0% de microenxertos com folhas expandidas. As avaliações histológicas evidenciaram maior espessura da região cortical em porta-enxertos de P. edulis, enquanto em P. setacea foi observado menor número de camadas corticais, maior quantidade de fibras floemáticas e de alcaloides, características que influenciaram negativamente o resultado da microenxertia.

This study aimed at evaluating the effect of Passiflora species used as rootstocks on the success and development of ex vitro interspecific micrografting, and at anatomically characterizing the region where it is done. Stem apices of Passiflora edulis f. flavicarpa were grafted onto the hypocotyl of P. alata, P. cincinnata, P. edulis, and P. setacea seedlings. The micrografting development, the occurrence of adventitious shoots in rootstocks and the percentage of micrografts with expanded leaves were evaluated. The rootstocks were subjected to anatomical and histochemical evaluation. Micrograft development and adventitious shoot number were higher when P. edulis rootstocks were used, and lower for rootstocks of P. setacea and P. alata. P. edulis, P. cincinnata, P. alata, and P. setacea rootstocks developed 30.3, 10, 1.6, and 0% of micrografts with expanded leaves, respectively. The histological evaluation revealed increased thickness of the cortical region in rootstocks of P. edulis, while in P. setacea a lower number of cortical layers, increased phloem fibers, and increased alkaloids were observed, characteristics that negatively influenced the micrografting outcome.

O objetivo deste trabalho foi avaliar diferentes meios de cultura, utilizados sobre o ponto da enxertia, na microenxertia ex vitro para a eliminação do Cowpea aphid-borne mosaic virus (CABMV), em plantas de maracujá-azedo (Passiflora edulis Sims f. flavicarpa Deg.). Ápices caulinares, provenientes de plantas infectadas, foram microenxertados em plântulas obtidas pela germinação de sementes em substrato comercial esterilizado. Foram conduzidos experimentos com a microenxertia realizada no hipocótilo e no epicótilo, e foram utilizados cinco meios de cultura, que diferiam na concentração de fitorreguladores, aplicados no local da enxertia. O índice médio de microenxertos com folha expandida foi de 27,22 e 32,22%, quando a microenxertia foi realizada no hipocótilo e no epicótilo, respectivamente. Na microenxertia realizada no hipocótilo, não houve efeito da aplicação de meios de cultura. Na microenxertia realizada no epicótilo, o meio MS acrescido de 0,1 mg L-1 de AIB e 1 mg L-1 de BAP proporcionou 53,3% de microenxertos com folha expandida, número superior aos demais tratamentos e maior desenvolvimento das brotações. A indexação realizada pelo teste ELISA indireto, 80 a 100 dias após a microenxertia, mostrou que 93% das plantas testadas não apresentavam vírus detectável.

The objective of this work was to evaluate the effect of different culture media, used over grafting spot, in ex vitro micrografting, in order to eliminate the Cowpea aphid-borne mosaic virus (CABMV) in sour passion fruit plants (Passiflora edulis Sims f. flavicarpa Deg.). Cauline apexes, originated from infected plants, were micrografted in seedlings obtained by the germination of seeds in commercial sterilized substrate. Experiments were conducted with the micrografting performed on the hypocotyl and epicotyl using five culture media, which differed on their concentration of phytoregulators applied on the grafting spot. The average rate of micrografting showing expanded leaf was 27.22 and 32.22%, when micrografting was performed on hypocotyl and epicotyl, respectively. In the micrografting performed on the hypocotyl, there was no effect of the culture media applied. In the micrografting performed on epicotyl, the MS medium supplemented with 0.1 mg L-1 3-indolbutiric acid (IBA) and 1 mg L-1 6-benzylaminopurine (BAP) resulted in 53.3% of micrografting showing expanded leaf, a number superior to the other treatments, and in a larger development of shoots. The indexing performed by the indirect ELISA test, within 80 to 100 days after micrografting, showed that 93% of the plants tested presented no detectable virus.
